Depends on your gears. 

I would use Deacon (Lead), Maulie 2nd to taunt everyone so you don't have to speed tune your team  Brago(buffer), and finally Rhazin to nuke. 

The only problem running this team is you'd need Deacon at at least 300 speed and 300+ accuracy. 

If you can't achieve that then going Gorgo + Deacon + Maulie + Rhazin seems like a good choice too. 

You have enough heroes to make several different types of teams

- Speed Nuking Team

- Resistance Team

- Stall Team

Forgive me, not sure what the mechanics of those teams are, is one more effective than the others?

Well - I don't think 1 team is better vs. another.

I think it all comes down to play styles.

Speed Nuking set ups = Fast & Furious 

- It tries to go first and kill the enemy in a single blow

- It seeks to do a 1 turn kill.

Resistance set ups = Refuses to Budge 

- It tries to go second and with stand all the enemies deadly assaults.

- Than when the moment is right the set up unleashes a deadly counter attack.

Stall Team set ups = Annoying & Painful

- This set up can go first or second depending on how you set up it.

- The set up doesn't try to win the fight.

- The set up tries to prevent itself from losing the fight.

- The set up tries to stall the fight out into a draw. 

In the process of going for a draw, The set up uses very annoying heroes.

The goal of the set up is to make the opponent so annoyed & upset.

The opponent leaves the battle out of frustration resulting in a victory for you.

Each Set up has a different play style.

Each Set up can be effective against different opponents you might run across in arena.

I, personally, have created several different arena set ups for myself.

I enjoy playing different play styles in arena for fun.

In Addition, It can be very beneficial to have different team play styles.

It can help you counter more teams in arena.

Obviously, I have been playing the game for a while.

I have a lot of different set ups which I have been working on over time.

You don't have the right heroes to create all the set ups I have.

However, I do think you could make a few set ups.

- Speed Nuking set up

- Resistance set up

It is something to think about.
